
Distinguishing the monosaccharide isomers in carbohydrates and natural products is the most elementary and important task. Traditionally, researchers using pre- or post-column derivation enhancedchromatographic and mass spectrometric methods to identify the type of monosaccharides in samples, which were time-consuming and slow.
Researchers in Prof. ZHOU’s team observed the formation of [2M + Na – H2O]+ of monosaccharides in the water/methanol (50:50, v/v) solution of NaCl. And the corresponding MS/MS information can be used for the rapid distinguishing of 8 common seen D-sugars isomers (Glc, Gal, Man, Fru, Rib, Ara, Xyl and Rha). The relative ratios of some of the product ion pairs yield unambiguous distinction. In addition, the use of PCA demonstrated the spatial resolution of the MS/MS data of these studied monosaccharides and the clustering with that of the glycoside sample.
The method is a rapid and efficient analysis method of monosaccharides identification, which showed the application potential to distinguish common monosaccharides in the field of natural products and beyond.
